Yes, many beamline groups at the APS use nexus, especially in the sense that nexus is HDF5+naming conventions.

From python, nexpy (https://github.com/nexpy/nexpy) is very useful for working with nexus data, either from plain python programs, or from its very nice GUI.  You can also sort of "roll your own" by using HDF5 + the nexus conventions.  That's probably not "recommended", but it can work in a pinch.

Yes, there is a nexus plugin for areadetector.  In addition, the HDF5 plugin follows (as far as I know) the nexus conventions, and can also be discovered and handled well by nexus tools.
